Efficacy of Graviola Seed Extract (Annona muricata: Annonaceae) on E-Cadherin Gene Regulation and Cytotoxicity in MDA-MB-231 Cell Lines

Background: The present study assessed for the cytotoxic effect of Graviola seed extract (GSE) in MDA-MB-231
and its influence on E-cadherin gene expression molecular level. Aim: The effect of the extract on E-cadherin gene
expression was studied by semi quantitative-PCR. Study designed to know the cancer regulation of E-cadherin
genes and toxicity in MDA-MB-231 using GSE. Methods and Materials: The study revealed a dose dependent
1.2 and 1.3 fold up-regulation of E-cadherin in MDA-MB 231 cell line which were tested at 20 and 40 μg/mL of
GSE, respectively. Further to check the cytotoxic activity of the GSE MTT assay was performed. Results: When
tested the cancer cell lines showed an IC50 value of 36.28 μg/mL with highest percentage inhibition of 94.7 while
Doxorubicin (positive control) showed an IC50 of 22.01 μg/mL with a highest inhibition of 84.01% at 100 µM
concentration. Conclusions: Since E-cadherin expression increased post-treatment with GSE, we can say that the
GSE acts as cytotoxic towards cancer cells which were further substantiated by the MTT assay.
