HLA class II expression levels and alloimmune risk

To assess HLA class II expression, the researchers exposed human blood-derived endothelial colony-forming cells to IFNγ. The expression levels of HLA-DR, HLA-DP and HLA-DQ varied between individuals but correlated with expression levels in other cell types (including antigen-presenting cells) from the same individual. HLA-DQ was the most variably and least frequently expressed HLA-II subclass. HLA genotype correlated with HLA-DQ expression levels and genotypes associated with high endothelial HLA-DQ expression were also associated with enhanced graft injury. In HLA-DR-matched patients, the histological microvascular inflammation score was highest in biopsy samples from recipients of kidney transplants with an HLA-DQ5 genotype.
